WBI’s models for Rheumatoid Arthritis (RA) cover all of the essential models for evaluation of anti-arthritic efficacy for small molecules, proteins and vaccines. Our scientists are experienced with evaluation of results for test materials and a wide variety of controls/reference materials used in hundreds of our prophylactic or therapeutic studies.

 

In addition to the basic read-outs for each model, optional analyses can be ordered for histopathology, cytokines, pharmacokinetic samples and collagen antibodies. 

 

 

A Final Report for each of our studies includes all details of materials, procedures, observations, measurements, data and statistical analysis for treatment efficacy.

 

 

 

Overview of Washington Biotechnology RA Models

 

 

 

Collagen-Induced Arthritis (CIA)        The “standard” animal model for evaluation of anti-arthritic activity

 

                                                             - Model based on immunization with bovine collagen to develop antibodies against bone and cartilage         

 

- 45-day study

 

- Species:     mouse or rat

 

- Prophylactic or Therapeutic

 

- Basic read-outs: Arthritic Index, body weights

 

- Optional:     Paw volumes, histopathology, cytokines, and collagen titers

 

 

Pristane-Induced Arthritis (PIA)         Non-immunogenic model that most closely simulates the autoimmune RA condition in humans

 

                                                            - 30-day study

 

- Well-defined inflammatory agent (pristane oil)

 

- Excellent model especially for the early stages of RA and hence for prophylactic or therapeutic intervention early in the disease

 

- Species:     rat

 

                                                            - Prophylactic or Therapeutic

 

- Readouts:   Arthritic score, body weights

 

- Optional:     Paw volumes, histopathology, cytokines, and collagen titers

 

 

Adjuvant-Induced Arthritis (AIA)       Economical in vivo screen; one of the longest established animal models for testing arthritis treatments

- Reliable model with less well-defined mechanism than CIA or PIA

Ovalbumin-Induced Arthritis (OIA)     Advantage of synovial fluid measures at convenient volumes

Air Pouch Model                                  Measurement of infiltrating cell types in response to inflammatory agent (carrageenan or other)

 

                                                                        - 6-day study

 

                                                            - Species:     mouse or rat

 

- The lining within the air pouch produced by injection of sterile air under the skin of the animal closely resembles the synovium in joints providing a convenient surface for study of the inflammatory cell response.                         

 

- Read-outs:  CBC, cell differentials, exudate volumes and cytokines

 

 

 

 

 PG-PS 10S Rat Model                        Allows evaluation of compound efficacy for acute or chronic (or both) inflammatory phases

 

                                                           - 22-day study

 

                                                            - Species:     rat

 

- An initial acute inflammatory response is seen within 3-6 days post-injection followed by a decline. A chronic response follows with a peak in measurements at approximately 12-16 days post-PG-PS 10S.
